Différenciation génétique de deux huîtres creuses Crassostrea gigas et Crassostrea angulata : apport des marqueurs microsatellites ArchiMer

Les définitions taxonomiques de Crassostrea angulata et Crassostrea gigas font l'objet de controverses. Les similitudes morphologiques et physiologiques ainsi que l'homogénéité des fréquences allozymiques entre populations des deux taxons orientent les auteurs vers un regroupement au sein d'une seule espèce. Récemment, une étude basée sur l'analyse par RFLP de l'ADN mitochondrial a révélé des différences permettant de distinguer C. angulata et C. gigas. The use of microsatellite markers for parentage analysis in the Pacific cupped oyster, Crassostrea gigas (Thunberg) ArchiMer

Highly polymorphic genetic markers can be useful tools for the analysis of parental contributions. The present paper reports 3 experiments using microsatellite markers to analyse parental contributions in controlled in vitro factorial crosses of the cupped oyster Crassostrea gigas. Parentage analysis was eased by the large polymorphism observed at the 3 loci studied. In a cross between 20 males and 20 females, a quarter of the progeny could be assigned to a given parent using a single microsatellite locus showing 28 alleles. In 3 crosses between 5 males and females, all the progeny could be assigned to a family by using 3 loci. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). DNA fragments ranging from 250 to 500 bp were size-selected by agarose gel electrophoresis and ligated into the dephosphorylated blunt-ended SmaI-linearised pBKS2 plasmid (Stratagene Cloning System, CA). Experimental examination of reproductive isolation is the first step in understanding li,hybridization processes. Here, we studied preferential fertilization between 2 cupped oyster taxa, Crassostrea angulata and Crassostrea gigas, as a potential prezygotic reproductive isolation. Early examination of sperm competition is now possible by molecular analysis of oyster embryos. This avoids the confounding effect of differential mortality during the larval stage. Soybean cultivars have been characterized mainly by morphological and biochemical traits. However, these traits have not been sufficient to characterize the large number of cultivars eligible to receive protection under the Brazilian Cultivar Protection Act. The allele frequencies of 10 microsatellite loci previously described for sheep as BM1314, BM6526, OarFCB128, OarHH64, OarCP20, OarHH47, OarFCB48, OarHH35, OarHH72 and BM2508 were estimated for the Uruguayan flocks. A representative sample of 101 individuals composed by the two predominant breeds (76% Corriedale and 24% Australian Merino) was used. Most laboratories have improved their exclusion probabilities (PE) by the addition of DNA microsatellite loci to standard blood-typing results. Comparative genetic structure in pines: evolutionary and conservation consequences RChHN

Pines have been the focus of several studies that estimate population genetic parameters using both allozymes and chloroplast single sequence repeats (SSRs). Also, the genus has also been recently studied using molecular systematics so that we now have a more clear understanding of their evolutionary history. Genetic differentiation in geographically close populations of the water rat Nectomys squamipes (Rodentia, Sigmodontinae) from the Brazilian Atlantic Forest Genet. Mol. Biol.

We examined the genetic structure and the effects of a bottleneck in populations of the water rat Nectomys squamipes, a primary host of Schistosoma mansoni. Eight microsatellite loci were studied in 7 populations from the Sumidouro region of the Brazilian state of Rio de Janeiro. Our data, covering a four-year period during which a bottleneck occurred, revealed substantial variation (6-31 alleles per locus) and high levels of both observed (0.718-0.789) and expected (0.748-0.832) heterozygosity. The isolation of (GA)n microsatellites using a highly microsatellite-enriched library is described for the first time in common bean (Phaseolus vulgaris L.). A relatively simple and effective method to isolate DNA repeats from microsatellite-enriched libraries based on hybridization-capture of repeat regions using biotin-conjugated oligonucleotids and non-radioactive colony hybridization was carried out. The objective of this work was to characterize mandarin (Citrus spp.) germplasm from Southern Brazil by morphological and molecular analyses. Thirty seven cultivars from 34 distinct mandarin varieties were evaluated by morphological and agronomic traits of leaves, flowers and fruits, and by microsatellite markers. The morphological and agronomic characteristics suggested that almost all varieties can be produced for commercial use, and some, as the Satsuma variety, are recommended for breeding programs.
